The three-year-old Lonhro colt, ridden by Sam Clipperton, was held up last of the five runners on the inside as his Godolphin stable companion, Holler, with Christian Reith up, took them along in the early stages.
Holler, racing wide, then settled in second, had every chance with two furlongs to go and took the lead briefly inside the final furlong before Exosphere, switched wide in the straight, swooped to win going away.
Exosphere, whose time came to 1m 10.62s on soft going, won by a length and a quarter from Press Statement, with Holler a further half length back in third.
John O'Shea said: "Exosphere unleashed a powerful finishing sprint for an emphatic win. He was very relaxed in the run which is important and sprinted well late.
"He has had a good blow. There is good improvement to come on whatever he did today and it was nice to see him come back from the adversity of his Golden Slipper run.
"I thought Holler's run was outstanding but his condition gave out over the last 100 metres which we thought might happen.
"He was dogged throughout the run and the horse that travelled on his inside in the first half dropped out which confirms how well he ran. I think he is still one run away from his peak."
Both Godolphin horses are being aimed at the Group One De Bortoli Wines Golden Rose over seven furlongs on September 12.
