Penwith goes after a first Graded win in the G2 Royal Delta Stakes over an extended mile on dirt at Gulfstream Park, USA, on Saturday, February 13.
The five-year-old mare returned from an 11-month layoff with an easy victory in a mile dirt allowance optional claimer at the same course on January 21.
The daughter of Bernardini has been placed in a couple of G2 races on dirt, including when third in the nine-furlong Demoiselle Stakes at Aqueduct in November, 2013, narrowly beaten by subsequent G1 winners Stopchargingmaria and Got Lucky.
Her trainer Kiaran McLaughlin said: "Penwith is doing well.
"She had been away a long time before her last run but she had been training great and we were very excited to get her started again.
"It is a little quick back but she is doing excellently and we are ready to go.
"We hope that she has a big year this year and gets some graded stakes.
"She's a five-year-old now but is lightly-raced and hopefully she can run in some big races."
Luis Saez, who last rode Penwith in 2014, resumes the partnership on Saturday when the Godolphin runner takes on five rivals, including Best Behavior and Sandiva, from stall four.
